[초청강연] Epigenomic and transcriptomic approaches to identifying cell fate determination

The mode of the spatial and temporal regulation of genes can be interpreted by analyzing the genetic and epigenetic features. The heritable epigenetic regulations without DNA sequence changes are usually mediated by various types of histone modifications, DNA methylation, and chromatin interactions. Thus the epigenetic mechanism is applied to explain many unsolved questions such as non-genic causes of cancer, fine-tuning of developmental program, and cell fate determination. text-indent: 21.3pt; line-font-family: Calibri, sans-serif;">Here, I am going to introduce our current approaches to understanding how epigenomic and transcriptomic dynamics are used to control various cellular activities in a cell-type specific manner. Using intestinal organoid and bladder assembloid models, we identified dynamic epigenetic regulation during stem cell differentiation and tumor phenotype changes, respectively. Comparative chromatin structure analysis of small and large intestinal stem cells enabled to discover key regulators from gene regulatory network. In addition, an integrative analysis of the transcriptomic and epigenomic changes confirmed cellular identity and plasticity. We expect our study could provide a valuable methodology to reveal how cells maintain their identity and control distinct transcriptional program.
