Pcz solutions, mutant screening

- Yesterday I cut the sections of the rice mutant for lamina assay. In case of phyB mutants some of seedlings have an exagerating lamina bending after growing in white light (which is the phenotype of those plants so is totally expected), but I tried to pick for the experminet the ones that doesnt present that level of bending, but are more similar to wildtype plants. But anyway in a separate vessel I put those with exagerate bending.
What I thought is that according with Prof. Park, the lamina bending of mutant phyB is due to BL synthesis, and if that assumption is correct is means that muntant plants will no present much difference with wild type planes after the treatment with pcz (the pcz should "revert" the phenotype). If that is the case, I should pick as an starting point in the experiment sections that dont present exagerate bending, because as we talked before, probably pcz is able to "prevent" the bending, but not "revert" the bending that was already in the plant....
Right?
... anyway today I will transfer the sections to the different pcz solutions...
- Since I have to prepare solutions for the lamina, I will prepare just a lot amount to do mutant screening of rice plants too.
- I will test with pUC19 construct the c-cells that we prepare yesterday with JiNyeong
