Yesterday,

I did RT revision and could see the band even the size marker. On later it came to know from the day before yesterday, the U-V in gel doc was out of order. So I could not continue the RT yesteday.

 Next I did gDNA extraction from F2 population of dw4 X gul4  and PCR with dCaps markers primer for gul4 mutation.

The F2 seeds were sown in Kan MS and only the Kan resistant seedlings were transplanted to soil,so theoretically all the seedlings transplanted in soil contain dwf4-1 mutation. So I tested the presence of gul4 mutation in those seedlings. The results shows the different patternsof the bands. Please find the results in attached file.

 Today I will do gDNA extraction and PCR for genotyping F2 of bri1-5Xgul4.
