Yesterday,

 I prepared the tissue powder of T2 of 35S:CYP83B1:YFP:HA in Ws-2 abckground and did western of them as well as T2 of 35S:CYP83B1:YFP:HA in gul4 backgrounds. I did finish staining with primary antibody HA and today I will continue with secondary antibody and detection.

I did agro colony PCR and 3 ml agro culture of 35S:gul4 cDNA:YFP:HA.

I would like to attach the ppt with photo of F1 phenotype of gul4 double mutant.

Today

 I will continue western blot.

PCR to confirm the transformation of CYP83B1promoter gDNA:GUS in gul4 and ws-2 background and will take photo.
