RT PCR

Hi Professor,

Today I will do RT PCR with rice and brachypodium samples. Yesterday I already did ACTIN for rice and brachy to test the cDNA, and apparently rice cDNA is ready to be tested and in case of brachypodium one of the samples didnt amplify. I have to check if was just that PCR or maybe I have to make cDNA synthesis again with that RNA.
I have also to check some brchyopodiium seeds that I have in cold for germination to prepare some other treatments for RNA extraction. If no I have to wait until friday of this week when the 10 days of stratification ends.

Finally I am attaching some picture of the lamina bending assay to JMJ703 mutant and wild type plant. What I see is that normally the mutant plant has more bending that wild type, even in mock conditions, and when adding epi-BL just increase a little that difference. But with pcz I can not see any relevant, because in mock conditions the plants dont show too much inclination, same with pcz were there is almost no inclination at all. But the problem is that there is no significant difference between the mock and pcz treatment, becuse both shows almost no inclination. I have to check if in other papers they obtain more inclination in mock conditions or is the same as me.
