Yesterday I did the following experiments.

1.Western Blot of BL treated Ws-2 and gul4.

2.  LR cloning of gul4 CDS in pEarlyGate 101-HA.

3. Miniprep of Entry of gul4 promoter construct and restriction enzyme reaction.

4.Seed sowing of F1 of double mutatns of gul4 with bes1-D, msg2-2D, yucca1-D, dwf7-1D.

5. Tried to use the available primers in our promer stock for genotyping the double mutants of gul4 and dwf4, but failed may be due to grreat differences in annealing temperature.

6. Transplantation of F2 of double mutants of gul4 with dwf4 and bri1-5.

Today's Plan

1. Continue Western.

2. Sequencing of Entry of gul4 promoter construct.

3. Seed harvesting of F1 of gul4 and atr4-1 and 4-2.

4. seed harvesting of of F1 od some double mutatns of gul4 and seed sowing.
